Hybridization on Strip


The Reverse Dot Blot is the technical procedure used in all of our products.

The procedure consists in three steps:

a) DNA extraction (reagents not included in the kit) ;

b) PCR Amplification (reagents included in the kit): the desired fragments of gene are simultaneously amplified by the polymerase chain reaction (PCR). During this PCR, a label (biotin) is added to the amplified DNA fragments.

c) Hybridization/detection (reagents included in the kit): test membranes bear covalently-linked DNA probes which specifically recognize fragments of each gene sequence, amplified during the PCR. There is one or more probes for the control of the developing process and a template as an aid for results interpretation.

During hybridization process, the probes specifically bind to DNA fragments amplified during PCR.As final result of the test, a band pattern is obtained which is interpreted with the aid of a control strip.
